When you encounter a Hunter (the big aliens with spikes on their back and a sword in their hand), the best way to kill it with one hit is by using the PISTOL (no other weapon will work).
Let him charge at you, dodge out the way and immediatley proceed toward its back. Press the R button (Fire) while aiming at the orange spot on it's back.
